What remains active inside the formulation?

Our research vision for preserving a selected protein ingredient’s useful function through formulation, handling and measurement. A defined question for a prospective qualified partner.

Collaboration vision · Subject to institutional discussion and agreement.

The ingredient should retain a reason to be there.

The proposed study asks whether a selected protein keeps its intended function through formulation, and whether the method can distinguish genuine loss.

We want to investigate what happens to a selected protein’s useful function when it becomes part of a skincare formulation. The commercial attraction is a preparation whose contribution can be explained and tested, rather than a protein name used to decorate an ingredient story. The question begins with the intended function and a suitable assay, then asks what mixing, formulation conditions and handling change. This is VARUNÉ Bio’s research vision for a prospective qualified partner. It is not an announced UCL collaboration, a developed cosmetic product or a result. A worthwhile first investigation would identify whether the intended function survives a defined preparation sequence and whether the method can distinguish real loss from an apparent change introduced by the formulation matrix.

Explanatory geometry · no simulated or measured performance

Look for the expertise that can separate those explanations.

Protein characterisation, assay design and process understanding could combine around one bounded compatibility question that a qualified team can investigate.

UCL’s independent Biochemical Engineering research describes biological-product characterisation, biocatalysis and process understanding; its ultra scale-down work considers how process environments affect materials (UCL, n.d.i; n.d.h). We see a possible scientific fit for a carefully bounded protein-formulation question. The expertise sought could combine functional assay design, protein characterisation, formulation compatibility and a realistic comparison plan. The team and resources would have to be agreed through the relevant institutional route. We would not assume that a capability described on a public page is available for our preparation. The purpose of the conversation is to decide which uncertainty needs specialist investigation and which cheaper preliminary check should precede a broader study.

Keeping identity, amount and activity on separate lines.

A trustworthy formulation comparison distinguishes what protein is present, how much is recovered and what function the selected assay measures.

A protein concentration result does not answer every question about a selected ingredient. We would want evidence of what is present, an appropriate measurement of how much is recovered and a function-specific observation under stated conditions. Smith and colleagues’ original protein-amount assay is useful analytical context; it does not make a colour signal a measurement of retained activity (Smith et al., 1985). A complex preparation may also contain other responding components. The proposed study would choose complementary measurements according to the candidate and question, rather than require every available instrument. If a nominally unchanged protein amount accompanies a weaker function, that difference deserves investigation. If recovery is incomplete, the report should first resolve what the assay actually sampled.

Stability is a family of questions.

Different stability endpoints can lead to different interpretations; the intended protein function should decide which comparison deserves primary attention.

Yu and Dalby (2018) compared several stability measures in external transketolase variants and found that the inferred interaction between mutations depended on the endpoint. Their work also measured activity retained after a defined exposure. We use that independent science to motivate an endpoint-specific investigation, not to select a protein or transfer their conditions into skincare. A thermal transition, an aggregation observation and a functional assay may answer different questions. Our preferred first endpoint would follow the intended ingredient function under a declared formulation comparison. A qualified team could then decide which structural observation helps explain it. The study should resist presenting a convenient thermal number as a complete account of whether the ingredient remains useful in the proposed base.

(Yu and Dalby, 2018)

Defining the denominator before calling it preserved.

A normalised activity value becomes useful when the reference, recovery and assay conditions remain visible alongside absolute measurements and variability.

The activity-retention ratio compares the same operational function before and after a declared condition. It requires a defensible positive reference, compatible assay conditions and appropriate correction for sample recovery or dilution. The external enzyme paper uses activity ratios; the actual Bio proposal would need a method qualified for the selected protein and matrix. A ratio above or below one should prompt interpretation, not be forced into a reassuring preservation story. The report would retain absolute activity, recovered amount and variability beside the normalised value. If the assay extracts the protein from the base, it answers activity after that extraction; it does not automatically show the function accessible while the ingredient remains inside the undisturbed formulation.

A negative corrected signal flags background domination; values above 100% require investigation, not automatic claims of improvement. Challenge the assay before trusting a formulation difference.

A base can change the analytical signal as well as the protein, making matrix controls essential before interpreting apparent preservation.

A coloured or optically complex base can change what an analytical method sees. Compton and Jones (1985) examined the chemistry and interference of a protein dye assay; that warning supports matrix-specific qualification rather than a universal claim that one kit will work. Our proposed controls could include a matched base without the selected protein, a known addition tested through the sample preparation, a suitable inactive comparator and a dilution series where it is informative. The exact set would depend on the functional assay. If the base changes substrate availability, background or the reaction conditions, a lower signal may not be irreversible protein damage. We would want the study to test that alternative before crediting a stabilising intervention or discarding a promising ingredient.

(Compton and Jones, 1985)

Compare the conditions a developer can actually change.

A small set of relevant formulation variables can expose a useful compatibility window without disclosing an enabling recipe or multiplying inconclusive experiments.

The proposed formulation study would choose a small set of meaningful variables: the medium’s acidity or ionic environment, the order of addition, a relevant handling exposure or a candidate protective preparation. Those are hypotheses to agree, not a disclosed recipe. A comparison should retain protein loading, preparation age and analytical treatment well enough that the endpoint can be interpreted. Independent batches and suitable reference preparations matter more than repeatedly reading one favourable sample. We would ask which conditions could make the candidate useful at a realistic ingredient level, and whether an established alternative already provides the same function. The most valuable result may be a narrower formulation window, a revised method or a decision that the proposed approach deserves no further development.

A possible first design would compare protein in its reference medium with protein in a defined base, alongside the same base without protein. Fresh and deliberately conditioned preparations could separate an immediate matrix effect from change during handling. The functional result and independently assessed recovery would be interpreted together. The actual conditions, replicate plan and acceptance boundary would be chosen by qualified investigators after the assay is demonstrated to be informative for that comparison.

One evidence package. Different reasons to use it.

The scientific contribution and development recommendation can share one evidence package while retaining separate claims, readers and disclosure needs.

For a prospective research team, the contribution could be an assay boundary, an explanation of matrix-dependent activity or a reproducible preparation comparison. For a formulation sponsor, the output could identify which compatibility question is resolved and which next experiment would justify continued ingredient development. We would envisage an agreed brief, raw observations, analysis with appropriate uncertainty and a concise recommendation. A scientific poster could display the hypothesis, primary comparison and supported findings without exposing protected compositions or enabling steps. A laboratory activity result would remain separate from consumer benefit, safety, preservation efficacy and a finished-product claim. Those would require their own qualified evidence. The proposed package should show the value of the research without stretching its endpoint into a marketing promise it cannot support.

A comparison worth making.

Define the preparation, the measurable difference and the decision the evidence could change.

A substantial research question needs a manageable centre.

A focused mechanism or measurement question can give a prospective researcher original work without an obligation to keep producing favourable product demonstrations.

A prospective doctoral contribution could investigate why the formulation changes the relationship between recoverable protein and measured function, or develop a comparison method that remains interpretable across declared matrices. We would want an original question with a clear experimental unit, credible method training and a feasible source of materials. UCL’s current Biochemical Engineering doctoral page is an official route for exploring the academic field; supervision, admission and funding remain separate institutional decisions (UCL, n.d.e). This vision is not a studentship advertisement. The programme should allow an investigator to report contrary evidence and develop understanding, rather than require an indefinite sequence of favourable product demonstrations. A focused method or mechanism can be academically valuable even if the selected formulation direction is eventually rejected.

(UCL, n.d.e)

Agree the question before opening the protected details.

The first non-confidential discussion should establish the intended function and decisive comparison before protected details, resources and institutional terms are agreed.

The first useful discussion can stay non-confidential: the intended protein-function class, the kind of formulation environment, the current comparator and the decision a successful assay would support. Material identity and provenance, qualified capacity, handling permissions, funded scope, publication and ownership terms would need agreement before a study. UCL’s data and IP policies provide institutional context for that process, not permission for Bio to use a facility or claim another party’s work (UCL, n.d.g; 2019). We would want a prospective partner to challenge the proposed endpoint and identify the minimum evidence needed to trust it. If that question earns agreement and resources, the next milestone would be a study that distinguishes a real retained function from an attractive but analytically unresolved ingredient story.

A prospective study worth shaping

Give an ingredient a functional comparison.

Total protein does not establish retained activity. A weaker signal may instead reflect matrix background, dilution, substrate response or a difference in the amount being compared.

Build a matched-loading study with a formulation blank, an untreated reference and a stress history relevant to the intended use. Add an orthogonal identity or concentration measurement to interpret an apparent functional change.

What a first scope
could produce.

  • A protein and matrix identity record
  • Blank and dilution controls
  • A matched-activity comparison and uncertainty plan
  • A retained-function candidate-selection brief

Bring a real question.

Bring the current benchmark, the unanswered scientific question, sample or data access, and the result that would change your next development decision. An analytical partner can contribute method expertise; a researcher can challenge the mechanism; a sponsor can define the application constraint.

A possible first commission would be a bounded feasibility, protocol-design or evidence-analysis package, followed by a separately resourced experiment where warranted. Scope, costs, delivery capacity, rights and institutional participation would be agreed before work began. These are proposed opportunities, not an assertion that a laboratory service or joint programme is already available.
