VARUNÉ Bio | Evidence Observatory

Look beyond
the headline result.

Change the comparison. Examine the conditions.
Find the next question worth testing.

Explore the evidence

A result becomes useful
in context.

Five core datasets, with twenty-three source rows, plus a new analysis of 165 published friction trial medians. Three published studies and one official chemical-property snapshot.

This first edition connects biochemical light, polar-associated polymers and formulation reference molecules. Each view makes the observation, its limitations and a proposed next experiment visible together.

These are reproducible public-data comparisons. They help formulate development questions; they do not establish the performance of a VARUNÉ Bio material or a finished product.

Triple-mutant reported efficiency relative to reference NanoLuc with CTZ. Switch to FMZ: the same variant falls below the reference.

The CTZ triple-mutant row reports turnover 40 s⁻¹, Km 0.77 µM and efficiency 70 s⁻¹ µM⁻¹. Dividing the first two reported means gives about 51.95, not 70. The published columns are retained pending clarification; exact magnitude should not be treated as independently resolved.

Why this matters to development.

An assay developer can see why a headline brightness claim is not enough to choose a system.

External purified-enzyme data at 37 °C in 100 mM potassium phosphate, pH 7.50. Ratios compare published means; no uncertainty for a ratio, individual replicates, finished-formulation behaviour or VARUNÉ Bio result is inferred. Catalytic efficiency is not a measurement of perceived brightness. Source-table clarification is required for the CTZ triple-mutant efficiency: its reported value differs from the quotient of the reported turnover and Km means.

A question worth testing.

The useful selection may be an enzyme–substrate pairing, chosen for the application, rather than an enzyme ranked alone.

Proposed next experiment. Agree the target application, substrate budget and operating window; compare the selected pairing in the intended matrix with appropriate controls, total photon output and signal persistence.

Why this matters to development.

A formulator or partner can ask for the material identity and batch comparability that make an evaluation interpretable.

Residue proportions do not establish the complete polymer sequence, purity of another preparation, biological function or a product specification. Functional crude-EPS results must not silently inherit this purified-EPS characterisation.

A question worth testing.

A comparable preparation needs a reproducible identity specification before a performance comparison can mean much.

Proposed next experiment. Specify source, preparation and purification; compare composition and molecular-size distribution across independent lots before assessing formulation behaviour.

Formulation | Published evidence, explored

Retention is a question about conditions.

What does one published water-retention endpoint support—and what would be needed to transfer it into a formulation?

75.79%

Starting water remaining after 72 hours
Mean ± 2.5 percentage points | n = 3

One reported endpoint. No invented curve between measurements.

Why this matters to development.

A development team gets a concrete reason to design the right comparison before paying for a scale-up or making a finished-product claim.

No time course or numerical comparator is reconstructed from the figure. This is not a skin-hydration, barrier or human-use outcome. The unresolved method wording prevents an exact reproduction claim.

A question worth testing.

A material may retain water in a defined laboratory test yet behave differently in a mixed formulation, deposited layer or humid environment.

Proposed next experiment. Resolve the paper’s retention-condition discrepancy first: Results/Figure 3c specify silica gel; the method also mentions 43% RH. Then pre-specify a gravimetric test with matched solids, starting water, environmental control, benchmark and independent preparations.

± 2.1 percentage points
At 10 mg/mL | n = 3

Superoxide anion

± 3 percentage points
At 10 mg/mL | n = 3

Why this matters to development.

A research partner can identify which test answers their application question instead of comparing unlike headline percentages.

Chemical assay percentages are not human protection, skin benefit, clinical efficacy or a comparison of overall radical-scavenging capacity. No unreported HA comparator values are inferred.

A question worth testing.

The relevant assay and matrix may matter more to a development decision than a broad antioxidant label.

Proposed next experiment. Choose the oxidative challenge relevant to the proposed material use, include matrix blanks and interference controls, and separate chemical response from biological activity.

Why this matters to development.

Formulators can inspect the source identity and compare a few relevant molecular properties without mistaking them for product performance.

These are computed small-molecule descriptors. Free GlcNAc is not the intact EPS polymer. TPSA and XLogP do not establish ingredient safety, absorption, regulatory suitability or cosmetic efficacy. Source: National Library of Medicine; National Institutes of Health; U.S. Department of Health and Human Services.

A question worth testing.

Descriptors are most useful as prompts for a matrix-specific compatibility and stability test, combined with actual material identity.

Proposed next experiment. Define concentration, solvent system, pH and storage conditions. Measure compatibility and stability using suitable controls; do not substitute a computed descriptor for the experiment.

The conditions change the comparison.

Viscosity, speed, load and contact geometry. Follow a published dataset into the question a formulation team would ask.

Film credits

What does the liquid
leave out?

A viscosity measurement describes the liquid. A friction experiment adds the contact and the movement.

The new tribology chapter reanalyses 165 published trial medians from eleven participants and fifteen liquids. Switch between viscosity and a dimensionless Hersey comparison to see how the interpretation changes when speed, force and geometry enter the model (Fehlberg et al., 2025).

The two reference fits use the same 77 observations from seven higher-viscosity liquids. Their descriptive fit does not establish prediction in new people, penetration into skin or the performance of a Bio ingredient. It gives a formulation scientist a more precise reason to ask which conditions belong in a comparison.

The published numbers describe behaviour. A deposited structure offers a different way to examine the reference system.

Closer, at the molecular scale

A specific reference.

Rotate a native view of deposited NanoLuc coordinates. This is a different system from the coastal plankton footage.

PDB 8AQ6 | Chain A | C-alpha traceRotate with the controls or drag

The molecular reference has an identity.

This simplified trace uses 171 unique C-alpha coordinates from chain A of PDB 8AQ6, associated with Nemergut and colleagues' published NanoLuc work. It displays a static deposited conformation; the motion rotates the view.

Inspect PDB 8AQ6 and its validation record

A structure sharpens the question.

The record supports a view of this specific external protein conformation. It does not show Bio's enzyme, a plankton luciferase, a reaction trajectory or behaviour inside a proposed film. Ligands, waters and side chains are omitted from this simplified trace.

The next material question

Which evidence would show that the chosen preparation retains useful activity in the intended architecture?

Native rendering of external deposited coordinates. Smoothing and tube thickness are display choices, not atomic dimensions. No molecular dynamics or original Bio experiment is represented.

Rotate and inspect a deposited reference structure. This is a molecular-coordinate view, not a simulation of the reaction or proof of our material architecture.

From a source
to a worthwhile study.

We organise the analysis around a development decision: which comparison would alter material selection, what evidence is missing and what a fair experiment would cost to learn.

New releases should add reproducible value, rather than a growing feed of incomparable numbers. Dataset expansion will depend on source rights, relevant questions and a repeatable refresh process.
