A publishable question needs a real comparison

A publishable question needs a real comparison.

A literature collection can reveal a worthwhile gap, but it is not an original experiment. This candidate asks what bounded analytical comparison could produce useful new evidence and a credible research contribution. Possible questions include whether preparation changes a marker response, whether a matrix distorts calibration or whether orthogonal methods resolve a disputed target state. The first decision is whether the comparison is scientifically justified, affordable and capable of being interpreted. A publication ambition should sharpen that question rather than encourage a large unfunded programme or a predetermined positive result that the study is expected to deliver.

Using external studies to locate the gap

Use external studies to locate the gap.

Pilolli et al. (2023; issue 2024), Monaci et al. (2020) and Xie et al. (2024) examine different preparation and recognition questions. Matuszewski, Constanzer and Chavez-Eng (2003) provide methodological context for separating matrix interference from recovery in another analyte class. The proposed study would identify which uncertainty remains unanswered for its specific material and conditions. It would not present an external result as original Bio evidence or call a changed matrix automatically novel. The gap must concern a comparison whose outcome could change scientific interpretation or a real development decision, with the inspected literature limitations retained.

Design around independent evidence

Design around independent evidence.

The variance hierarchy helps distinguish material lots, preparation units and repeated readings under explicit assumptions. It is an illustrative design model, not a Bio dataset. A proposed study should replicate the unit relevant to its claim and keep pairing, randomisation and analysis compatible with the actual design. If resources allow only a narrow pilot, the conclusion should remain narrow. A large number of detector readings cannot replace independent materials or preparation. The analysis plan should preserve uncertainty and specify how ambiguous, failed or below-range results will be handled before the team sees which interpretation looks most attractive.

Nested variance decomposition
σtotal2=σlot2+σprep2+σread2\sigma^2_{\mathrm{total}}=\sigma^2_{\mathrm{lot}}+\sigma^2_{\mathrm{prep}}+\sigma^2_{\mathrm{read}}

A proposed additive random-effects model separates independent material lots, preparations and repeated readings.

σtotal2\sigma^2_{\mathrm{total}}
Total variance on the chosen response scaleresponse units²
σlot2\sigma^2_{\mathrm{lot}}
Between-lot variance componentresponse units²
σprep2\sigma^2_{\mathrm{prep}}
Between-preparation variance within lotresponse units²
σread2\sigma^2_{\mathrm{read}}
Instrument reading variance within preparationresponse units²

Model conditions. Independent zero-mean random effects and appropriate nesting. Not valid without sufficient replicated levels or under unexplained correlated effects. Log-scale analyses require interpreting variance on that scale.

The comparator before the story

Choose the comparator before the story.

A scientifically useful comparator should test the disputed mechanism and use a justified measurand basis. It may be a different preparation, authenticated reference material or orthogonal analytical method. Its suitability must be established for the question rather than assumed from instrument prestige. A proposed comparison should state what each possible outcome would mean and what remains unresolved. Agreement may support a limited inference; disagreement may identify an important gap. The study should not be structured so that one favoured platform must win. A meaningful negative or contradictory result can be a stronger contribution than a weakly controlled positive demonstration.

The defined comparison

SE(Yˉ)=σb2/B+σe2/(BR)SE(\bar Y)=\sqrt{\sigma_b^2/B+\sigma_e^2/(BR)}

Balanced random-intercept scenario with between-preparation SD 10 response AU and within-preparation SD 6 response AU. Preparations are independent; residual readings are independent with common variance. The naive estimate treats all BR readings as independent and ignores clustering. The 3D groups depict a design count, not measured sample values. No confidence interval, power or precision guarantee is calculated. Drift, pairing, operators, serial correlation and unequal group sizes need additional structure.

Preserve provenance and versions

Preserve provenance and versions.

Original observations would need sample identity, preparation lineage, acquisition conditions, calibration version and analysis code or calculation record where relevant. Changes to exclusions, models or reporting units should be retained rather than silently overwrite the initial plan. A future evidence collection could make these relationships easier to inspect, but the interface itself is not scientific proof. The study report should allow a qualified reviewer to trace each interpreted value to its source and assumptions. That makes the work useful for a development buyer and creates the basis for transparent scholarly reporting if the eventual evidence warrants it.

Contribution and publication are distinct commitments

Contribution and publication are distinct commitments.

Experimental delivery, analysis, writing and funding are separate contributions. A prospective study should resolve responsibilities, data rights and publication expectations before work, with authorship based on actual scholarly contribution. Paying for a scope does not automatically establish authorship or ownership of all resulting knowledge. This public proposition makes no journal submission, acceptance or partner claim. It also does not expose enabling confidential methods. The eventual publication route would depend on the strength and completeness of the evidence, an appropriate article type and permission to release the relevant material and results.

Fund the smallest complete study

Fund the smallest complete study.

The proposed first scope should be small enough to complete coherently and strong enough to interpret. That includes reference access, qualified laboratory resources, statistical planning, analysis and reporting—not merely the visible instrument run. Current quotations and a responsible lead are prerequisites. If the material cannot be authenticated or the comparator cannot resolve the question, the study should be revised before spending. A pilot may justify a later original comparison without being misrepresented as a finished publication dataset. The aim is to earn an evidence milestone through actual delivery, with a clear stop or pivot if the hypothesis does not survive.

The next proposed comparison.

A study record ready for critical review

A study record ready for critical review.

The proposed output would be a versioned study specification and, after qualified funded delivery, a report preserving individual observations, uncertainty, deviations and limitations. A research partner could judge whether the question merits collaboration; a commercial sponsor could assess whether the evidence is worth funding. The record could support a publication decision only when original work exists and the scientific contribution is defensible. External papers currently support the design direction, not completed Bio results. The next conversation should identify the comparison, independent units and decision value, with resources and release rights agreed before the study is represented as active.
