Identifying the marker. Understand the matrix.
Can a named milk-protein marker be recovered and recognised in a defined dark-chocolate preparation?

A deliberately bounded question
A deliberately bounded question.
Dark chocolate is an attractive analytical challenge because a convincing signal must survive a complicated preparation. It is also a setting where loose language can do harm. This candidate asks whether a specified milk-protein marker can be investigated in a defined dark-chocolate matrix using qualified analytical comparators. It does not offer a food-safety test, consumer advice or an all-milk detection claim. The useful first decision is whether the proposed marker and preparation warrant a separately funded feasibility experiment, before committing to a bespoke sensor or a broader commercial development programme.

Casein, β-lactoglobulin and peptides derived from those proteins answer different questions. The presence of a whey marker cannot establish the abundance of every milk-derived material, and its absence cannot exclude all milk proteins. A research brief would name the target, its relevant processed states and the intended reporting unit. Xie et al. (2024) illustrate how antibody recognition depends on a particular epitope; Monaci et al. (2020) examine processing-dependent peptide measurements in cookies. These are reasons to specify the target state, not evidence that any particular binder works in our proposed chocolate preparation.
Preserve the mass balance
Preserve the mass balance.
A signal measured in a diluted extract is several steps away from a mass fraction in the original portion. The proposed analytical record would retain sample mass, extraction volume, aliquot changes and net dilution. If digestion generates a peptide marker, converting its amount to parent protein or total milk protein requires an independently justified factor. Pilolli et al. (2023; issue 2024) treat that conversion explicitly. The simple extract mass balance shown here applies only to the named extracted marker; it contains no hidden assumption that extraction is complete or that a marker represents whole milk.
Unit-consistent mass balance for the named extracted marker; conversion to total milk protein requires a separate evidenced factor.

- Marker concentration in the measured diluted extractµg/mL
- Total extraction volumemL

Model conditions. No unrecorded concentration or aliquot factor. Extracted-marker result is not automatically whole-protein or total-milk content. Preparation loss is not silently corrected.

Making preparation an experimental variable
Make preparation an experimental variable.
Cocoa solids, lipids and polyphenol-rich components make preparation choices consequential. A candidate study should compare feasible extraction and clean-up arms against the same representative sample, while tracking whether the process preserves the intended recognition site. A harsh extraction useful for mass spectrometry may not be compatible with a binding assay. The fairest comparison therefore need not feed one identical extract into every detector; it must instead document each method’s preparation and determine whether both report the same measurand. Otherwise a preparation difference can masquerade as a detector advantage.
Examine the comparison
| Candidate marker basis | Useful analytical question | Limit |
|---|---|---|
| β-lactoglobulin recognition | Is the relevant recognition state accessible? | Cannot exclude all milk-derived proteins |
| Casein-related marker | Can the selected casein basis be measured? | Needs state and preparation definition |
| Digestion-derived peptide | Is the named sequence marker recovered? | Requires justified conversion before parent/total-protein reporting |

The defined comparison
Hypothetical concentration results on the same original-sample-equivalent and dilution basis, with 10 μg L⁻¹ added. Negative increments or recovery above 100% are retained to expose background, interference or uncertainty. A fresh spike need not behave like a processed, incurred or bound marker; pre- and post-extraction spikes test different effects. No automatic recovery correction, assay validation, allergen-safety conclusion or selected analyte is represented. Vessel heights are schematic; the graph is the quantitative comparison.

Separate spike performance from incurred material
Separate spike performance from incurred material.
Adding purified target to a finished extract is a useful first challenge to detection. Adding it before extraction probes a larger part of the chain. Neither automatically reproduces material incorporated before chocolate processing. The proposed feasibility design would label these arms separately and retain their limited conclusions. Pilolli et al. (2023; issue 2024) use characterised incurred chocolate; Monaci et al. (2020) use an incurred bakery comparison. Those external designs guide the questions to ask. They do not supply representative Bio samples, a validated preparation or numerical acceptance limits for a new study.
An orthogonal comparison with a purpose
An orthogonal comparison with a purpose.
An immunological response and a peptide mass-spectrometric response could help identify different reasons for failure. Agreement is useful only after units, marker identity and preparation basis have been reconciled. Disagreement should trigger an investigation of target state, recovery and calibration, rather than a claim that the more exciting sensor is correct. A qualified laboratory would need to propose the actual comparator method and confirm its suitability for the submitted matrix. Research-stage method comparison should remain distinct from routine food testing, and the food business must retain its established safety decisions.
A separate resource gate
A separate resource gate.
This branch should have a named analytical lead, a defined matrix owner, suitable standards, agreed data rights and a budget independent of the flagship materials work. The first gate would ask whether blank and positive-control behaviour can be interpreted at all. The next would ask whether representative preparation and comparator access are achievable. A poor recovery result might justify changing the marker or stopping the branch. No launch date, contracted laboratory, customer or funding award is implied by this candidate page; the proposal exists to make a qualified funded discussion concrete.

The proposed deliverable would describe the tested preparation classes, target identity, comparator logic, calibration basis and unresolved matrix effects. It would identify which observation changes the next development decision and which claims remain outside the study. A potential assay developer could then decide whether to fund a specific recognition experiment; a research partner could judge whether the analytical question merits its resources. Any original results would be reported with their actual uncertainty and independent units. A successful first feasibility check would earn a next study, not a food-safety certification or a market-ready assay.
