Replacement is a different question from association
Ask whether a new measurement can replace the existing method, keeping that practical question separate from whether their readings correlate.
A new method may track the same trend as an existing method while giving results too different to use interchangeably. The proposed benchmark record begins with the intended action: replace a protocol, combine datasets or use a transformed measurement in an existing acceptance rule. It asks how much disagreement would matter to that action and which paired observations can assess it. A high correlation is not the answer by itself. A formulation or assay buyer needs a comparison that addresses the actual workflow, with the measured range and practical tolerances defined before the new method is presented as equivalent or ready for routine use.

Difference and magnitude reveal another view
Inspect paired differences across magnitude to reveal disagreement that a strong correlation can conceal in a proposed method comparison.
Bland and Altman (1986) describe difference-versus-mean assessment for paired methods and explain why correlation can conceal disagreement. The author-hosted corrected reproduction was inspected as a method source; its clinical example data are not redistributed or used to claim material performance. The proposed formulation application would retain the two measured values, their difference and their mean, then inspect bias, scale dependence and unusual observations. This gives the buyer a direct view of how a substituted method could change a decision rather than rewarding the new method merely for reproducing the ordering of specimens.
Inspect method difference against the average measured magnitude.

An agreement interval needs its assumptions
Check distribution and repeat assumptions before interpreting an agreement interval, then compare it with the application’s separately defined tolerance.
A summary range of differences requires a suitable distribution, stable behaviour across the measured range and an appropriate repeat design. It is not an application tolerance automatically supplied by the data. The proposed record would distinguish estimated agreement behaviour from the buyer’s acceptable difference and retain uncertainty in the estimate. If variability changes with magnitude, a simple constant interval may be inadequate. The team would need to inspect that structure rather than applying a familiar diagram mechanically. The method comparison should answer whether the new approach is useful for the stated decision, not whether its plot resembles a textbook example.

Repeated pairs cannot be counted as independent specimens
Retain specimen and session identities when assessing repeated pairs so that additional readings do not become additional independent materials.
Several paired readings on one specimen can help assess repeatability while still sharing the underlying unit. Bland and Altman (1994) explain the dependence problem in repeated data. The proposed method-comparison plan would record specimen, preparation, session and pairing identifiers, then specify an analysis appropriate to that design. A large number of repeated readings would not become an independent replacement-method sample size. This matters when a buyer considers using a faster protocol across production lots: the validation question may require independent materials and an operating range broader than a carefully repeated measurement of one laboratory specimen.

Points are discrete design calculations, joined for comparison. No experimental observations or confidence intervals are shown.
Fund the preparation or repeat that addresses the uncertainty relevant to the development decision.
The defined comparison
Balanced random-intercept scenario with between-preparation SD 10 response AU and within-preparation SD 6 response AU. Preparations are independent; residual readings are independent with common variance. The naive estimate treats all BR readings as independent and ignores clustering. The 3D groups depict a design count, not measured sample values. No confidence interval, power or precision guarantee is calculated. Drift, pairing, operators, serial correlation and unequal group sizes need additional structure.
Inspect the calculated values

Calibration and agreement have different jobs
Distinguish traceable calibration from agreement between methods, identifying which measurement question the buyer actually needs the study to answer.
Calibration against a justified reference quantity can address a different question from agreement between two imperfect methods. The proposed record would retain any calibration transformation and its uncertainty, as well as the paired method outputs. JCGM’s measurement framework makes the input model and uncertainty relevant to the resulting quantity (JCGM, 2008). A favourable agreement view would not establish absolute accuracy without an appropriate reference. A buyer can then decide whether its workflow needs reliable substitution, traceable absolute values or both, and avoid claiming that one method-comparison study has answered every metrology question associated with the new approach.
A benchmark should reflect the receiving workflow
Cover the specimen types and operating conditions of the receiving workflow before interpreting a narrow laboratory benchmark as general substitution evidence.
The paired observations should cover the specimen types and conditions in which the buyer would use the new method. A narrow laboratory range may be adequate for an early feasibility decision while leaving broader replacement claims untested. The proposed plan would identify relevant extremes, preparation differences and measurement limits before selecting the sample set. It would also compare the practical burden of each method, including required sample handling and information inputs. A cheaper or faster approach is worthwhile only if its measurement behaviour is sufficiently useful for the application; neither benefit is assumed or quantified without a defined operational comparison.
A method-replacement decision package
Deliver the pairing specification and acceptance rationale together so that a partner can assess whether independent replacement testing is justified.
The proposed deliverable would contain the paired endpoint specification, source context, acceptable-difference rationale and a comparison analysis plan. It would separate completed numerical checks from proposed independent validation and identify any remaining calibration requirement. A partner could use that package to decide whether a new protocol, instrument route or data transformation deserves a qualified study. Practical access, rights and delivery capacity would require agreement. The service proposition is a defensible measurement decision, not a certification that Bio has validated a replacement method or demonstrated commercial performance across a customer’s workflow.
The next proposed comparison.
A paired measurement specification with pre-defined acceptable differences
Let disagreement identify the next test
Let the pattern of disagreement select the next calibration, specimen or operating-range test, turning a failed comparison into a useful action.
If differences increase with magnitude, the next study may need a revised transformation or a narrower operating range. If one preparation type behaves differently, the method’s scope may need to exclude it or establish a separate calibration. If repeatability is poor, collecting more distinct specimens before stabilising the protocol may be wasteful. The proposed record would connect each finding to that concrete action and preserve the stop criterion. A buyer should leave knowing what would justify substitution or continued parallel testing, with enough evidence context to challenge the conclusion rather than accepting a high correlation as proof of agreement.
