What the formula contains is not where it resides.
A proposed phase-availability study brief connecting nominal preservative loading with the preparation actually exposed to a microbial comparison.
Proposed programme | Study scope, qualified delivery and funding to be agreed.
Where the material goes?
Phase location creates an analytical question.
A formula lists a total loading, but a dissolved ingredient can distribute between phases, associate with interfaces or interact with other components. An emulsion revision may therefore create a chemical question before it creates an interpretable microbial one. Bio’s proposed brief would identify which phase and species matter to the intended comparison. The initial objective would be a plausible mass balance and a measurement request, not an efficacy forecast. A sponsor could use it to decide whether a changed emulsifier, oil fraction or polymer warrants analytical characterisation before commissioning a larger preservation comparison.
Examine the development decision
What the formula contains is not where it resides.
Open the sequence to examine what a development decision needs.
Identify the exact preparation, its source, and the properties a second laboratory must be able to reproduce.
Explanatory geometry · no simulated or measured performance
Describe a partitioning model without pretending it is the formula.
Model simplification should be explicit and inspectable.
A simple two-phase equilibrium model can organise assumptions about the distribution of a species. It leaves out micelles, binding, interfacial adsorption and kinetic exchange unless those are explicitly added. The proposed record would state whether the candidate coefficient is measured, sourced from another preparation or merely assumed for exploration. It would also identify the actual species being modelled. An ionisable acid can require a different treatment from a neutral compound. The useful output is a list of questions that can be checked experimentally, rather than a single calculated concentration presented as the amount available to every organism.
Preserve the identity of the measured fraction.
Sample separation can alter the measured fraction.
Separating a phase for chemical analysis can itself change equilibrium or lose material on a filter, tube or extraction medium. A measurement request should define the sample preparation and recovery controls. It should distinguish total concentration from the operationally measured fraction. Bio could map these distinctions for an application scientist before analytical work is quoted. The reported method and recovery uncertainty would remain beside the result. A visually convincing phase diagram must not imply that a chemical fraction has been measured when it has only been modelled, or that one analytical preparation represents every microscopic environment in the finished product.
The formula changes the observation.
Investigate the relationship between formulation, phase availability, sample recovery and interpretable microbial evidence.
Making the mass balance auditable.
Identify missing volumes and species information.
The equation below derives aqueous concentration from total amount and a specified equilibrium partition ratio in a deliberately simplified two-phase system. Every quantity must share a compatible unit basis. The relationship is useful because it shows which volume and coefficient information is missing. It does not predict microbial exposure at interfaces, preservation performance or skin effects. A model experience could let the reader vary hypothetical phase volumes and inspect the assumptions. The decision would then be whether obtaining an actual distribution measurement is likely to alter the study design, rather than whether the calculated output looks commercially attractive.
An independent mass-balance derivation used to frame a measurement; no partition coefficient or efficacy data are invented.

Model conditions. Two equilibrium phases with additive volumes. No binding, micelles, interfaces or losses. Coefficient must fit the species, matrix and temperature. Aqueous volume must be positive; oil volume and the specified partition ratio are non-negative.
Change the conditions · examine the reference
The total is not the available fraction.
Change pH and phase volume. See how a defined neutral species can redistribute even when total loading stays constant.
The data, equations and controls remain available below if 3D cannot load.
Defined model · illustrative conditions
Move a control to compare the selected condition with the defined reference curve.
Separate total loading from phase-specific concentration before defining a functional comparison.
The defined comparison
Hypothetical monoprotic weak acid: pKₐ = 4.8, neutral-species partition ratio K = [HA]oil/[HA]water = 10, total 1 mmol per litre of whole formulation. Only neutral HA partitions into oil; phase volumes are additive and activities ideal. No micelles, adsorption, binding or non-equilibrium behaviour. Phase shares refer to solute mass, not liquid volume. This reference does not predict preservation efficacy, microbial safety or delivery to skin.
Inspect the calculated values
Calculation grid, not measured observations. Inputs and assumptions are stated above.

Control pH when the species can change.
Speciation can change alongside phase composition.
IUPAC’s activity-based pH definition gives the chemical basis for asking how an ionisable component is represented (IUPAC, 2025). A proposed study would retain pH and measurement conditions alongside the phase comparison. If the revision changes both oil fraction and acid speciation, a difference cannot be attributed to partitioning alone. A qualified analytical reviewer could advise whether the chosen species and approximation are appropriate. The brief would identify the simplest matched comparison that isolates the uncertainty. It would not supply a recipe or encourage a concentration adjustment outside a separate formulation and suitability assessment.
Connecting chemical availability to a separate biological observation.
Join chemical and biological observations through preparation.
An analytical fraction and a microbial count answer different questions. FDA BAM provides a cosmetics analytical context for recovery and enumeration, while ISO 11930’s public record identifies preservation-evaluation scope (Huang et al., 2024; ISO, 2019). A proposed project could connect the two through matched preparations, but should retain their distinct methods and uncertainties. The laboratory would define the microbiological study and suitability controls. An association between measured fraction and response would need enough independent preparations to interpret credibly. The evidence ledger should show that the chemical explanation remains a hypothesis unless the comparison actually supports it.
Giving the buyer an actionable analytical question.
Make the next measurement worth commissioning.
The proposed deliverable could contain a phase map, a assumptions register, a mass-balance worksheet and a quotation-ready analytical brief. A formulator revising an emulsifier system could use it to choose a first measurement. An ingredient supplier could use it to identify what additional evidence would make an application dossier more informative. Any later method development or microbial testing would be separately resourced with qualified partners. The value proposition is the resolution of a specific uncertainty that blocks a development decision. It would not claim that a partitioning model replaces preservation testing or that Bio already offers validated phase analytics.
The next proposed comparison.
Phase identity, mass-balance and comparative measurement brief
The revision that needs explanation.
Follow the specific revision that needs explanation.
An opening discussion should state the matrix type, ingredient class, changed phase or interface and practical constraint. A confidential formula can be shared later if needed under agreed terms. The first milestone would be a reviewer-approved mass-balance and measurement question that distinguishes total loading from the fraction of interest. If the simple model is inappropriate because binding or exchange dominates, the useful conclusion is to revise the framework. That can stop an unnecessary simulation or focus the next experiment. A credible commercial stream grows from those decisions and measured preparations, not from universal claims about where every preservative resides.
