The total is not the available fraction.

Investigate how phases, micelles and membranes distribute an ingredient before attributing behaviour to its nominal concentration.

Proposed programme | Application, materials, capacity and rights to be qualified.

Count the places a molecule can occupy.

A formula’s total concentration can be distributed between water, oil, interfaces, micelles and carrier membranes.

A formula’s total concentration can be distributed between water, oil, interfaces, micelles and carrier membranes. The fraction relevant to an observed response may differ from the nominal amount on the ingredient sheet. A formulator deciding whether to change an oil or surfactant needs to know which distribution matters to the development question.

The proposed investigation treats phase distribution as a testable hypothesis. It might explain a change in release, extraction or chemical exposure. The first task is to identify the possible compartments and an analytical operation that can resolve them without creating a new equilibrium that obscures the original formulation.

Fugit and Anderson (2014) investigated membrane binding and release in liposomes, showing that partition behaviour and transport need to be considered together. Their study is an external pharmaceutical example; its coefficients belong to the tested compound, lipid system and experimental conditions.

Liu et al. (2009) examined free benzalkonium chloride in micellar systems and reported dependence on surfactant and medium. That abstract-level evidence supports the importance of free-fraction measurement, not a preservative recommendation for a Bio product. An application brief would recover the relevant phase and assay information before considering whether either study provides an appropriate method analogy.

A deliberately simple balance.

For an ideal two-phase equilibrium, total active mass can be expressed as aqueous concentration multiplied by water volume plus oil concentration multiplied by oil volume.

For an ideal two-phase equilibrium, total active mass can be expressed as aqueous concentration multiplied by water volume plus oil concentration multiplied by oil volume. Introducing an oil-to-water concentration ratio gives the displayed estimate for aqueous concentration. It is useful because every assumption is visible.

The model excludes micelles, interfacial adsorption, ionisation, precipitation and concentration-dependent binding. Many real formulations contain those features. A reader could change phase volumes and a stated partition coefficient to understand why the same total loading can imply a different free concentration, while the page explicitly keeps that illustration separate from a validated prediction for the finished formula.

Whether the assay changes the answer?

Ultrafiltration can remove carriers but also bind active; dilution can change micellar populations; centrifugation may leave smaller entities in the sampled fraction.

Ultrafiltration can remove carriers but also bind active; dilution can change micellar populations; centrifugation may leave smaller entities in the sampled fraction. The proposed method record would state what each operation excludes and include active-recovery controls. An apparently free fraction is operationally defined until independent checks support the assignment.

Wallace et al. (2012) identify incomplete particle separation as a source of inaccurate encapsulation and release estimates. That warning applies to the interpretation of phase fractions. The report would preserve the separation history and distinguish a measured filtrate concentration from a claim about chemical activity at an interface or biological site.

[HA]w=CT(1−ϕo)(1+10pH−pKa)+Kϕo[HA]_w=\frac{C_T}{(1-\phi_o)(1+10^{pH-pK_a})+K\phi_o}

Hypothetical monoprotic weak acid: pKₐ = 4.8, neutral-species partition ratio K = [HA]oil/[HA]water = 10, total 1 mmol per litre of whole formulation. Only neutral HA partitions into oil; phase volumes are additive and activities ideal. No micelles, adsorption, binding or non-equilibrium behaviour. Phase shares refer to solute mass, not liquid volume. This reference does not predict preservation efficacy, microbial safety or delivery to skin.

Keeping pH and association in the same map.

For an ionisable compound, changing pH can alter both partition behaviour and solubility.

For an ionisable compound, changing pH can alter both partition behaviour and solubility. A pH adjustment might also change carrier surface interactions. The proposed comparison would therefore record pH, buffer composition and phase volumes together rather than treating a supplier partition coefficient as a universal material constant.

Luengo et al. (2021) investigated a local-pH mechanism in a PLGA system. That provides a useful counterexample to the idea that all delivery changes arise from physical transport of a carrier. The buyer’s active requires its own chemistry and measurements; the proposed programme would investigate competing explanations before recommending a more elaborate architecture.

(Luengo et al, 2021)

Connecting distribution to the decision.

A cosmetic development team may need the active to remain chemically protected in one phase during storage but become available during application.

A cosmetic development team may need the active to remain chemically protected in one phase during storage but become available during application. That is a trade-off between retention and availability, with different relevant conditions. The optimal total loading cannot be chosen independently of phase distribution and the function being investigated.

The proposed paid output is a compartment map, a method-readiness record and a bounded comparison of the preparation variables that can change the next decision. A later formulation evaluation could investigate whether the intended distribution persists after incorporation. No partition model alone establishes safety, skin efficacy, preservation or an available Bio delivery technology.

The phase changes that matter.

An opening discussion should identify the active grade, oils, surfactants, pH range, final base and current analytical approach.

An opening discussion should identify the active grade, oils, surfactants, pH range, final base and current analytical approach. The most useful first check may be recovery of free active through the proposed separation method, or solubility across a realistic phase-composition change. That check can determine whether a more complex study is warranted.

A meaningful milestone would be a reproducible phase-accounting observation that changes a formulation choice. If the method cannot resolve the needed fraction, the deliverable should say so and identify a qualified alternative. The commercial invitation is a precise decision about distribution and its consequence, not an unexplained ranking of delivery systems.
